细胞铁死亡（Ferroptosis）是一种铁依赖性脂质过氧化物积累所引发的新型细胞死亡方式。铁离子极大地加速了脂肪酸的过氧化反应，脂质过氧化物在铁的调控下累计，这些脂质reactive oxygen species（ROS）超出细胞抗氧化系统的承受能力，氧化过程损伤胞内蛋白、核酸和脂质，导致细胞死亡。

常规细胞凋亡或坏死常常伴随着细胞器肿大、染色质聚合、细胞膜完整性下降等特征，而细胞铁死亡在细胞形态上引起的改变甚微，往往只伴随细胞体积略微减小、线粒体膜密度增加等。

引发铁死亡通路（ferroptotic pathway）关键因素：

★ 贮藏在磷脂膜中的多不饱和长链脂肪酸（PUFAs）
★ 可氧化还原的铁离子
★ 有缺陷的脂质过氧化修复系统

在癌症研究领域中，Ferroptosis suppressor protein 1（FSP1）和 Glutathione peroxidase 4（GPX4）已成为重要研究靶点。FSP1和GPX4作为细胞铁死亡通路的负调节因子，阻碍细胞铁死亡。其FSP1和GPX4抑制剂则能有效激活癌细胞的铁死亡，该类抑制剂的药物筛选也已成为研究新型抗癌治疗方法的热点项目。
