BD Horizon™ Brilliant Stain Buffer Plus

Either BD Horizon Brilliant Stain Buffer or BD Horizon Brilliant Stain Buffer Plus should be used in multicolor immunofluorescent staining and flow cytometric analysis when using two or more BD Horizon Brilliant reagents. Whole human blood was stained with BD Horizon™ BV421 Mouse Anti-Human CD4 and BD Horizon™ BV711 Mouse Anti-Human CD19 (Top Plots) antibodies or BD Horizon™ BUV395 Mouse Anti-Human CD4 and BD Horizon™ BV605 Mouse Anti-Human CD19 (Bottom Plots) antibodies. Erythrocytes were lysed with BD FACS Lysing Solution (Cat. No. 349202). Two-color flow cytometric contour plots showing the correlated expression of CD4 versus CD19 were derived from gated events with the forward and side light-scatter characteristics of intact lymphocytes. Staining cells in the presence of BD Horizon Brilliant Stain Buffer (Cat. No. 563794/566349; Middle Plots) or BD Horizon Brilliant Stain Buffer Plus (Cat. No. 563795/568264; Right Plots) restores the major lymphocyte populations to their expected fluorescent staining patterns (dotted lines) when compared with cells stained without a Brilliant Stain Buffer (Left Plots). Flow cytometric analysis was performed using a BD LSRFortessa™ Cell Analyzer System.
