    Forkhead 转录因子家族参与横纹肌肉瘤和急性白血病的发生 (1-3)。这个家族中，三个成员（FoxO1、FoxO4、 FoxO3a）与线虫的同源基因 DAF-16 具有序列相似度，而 DAF-16 信号转导包括 IGFR1，PI3K 和 Akt 在内的信号通路 (4-6)。活化的 forkhead 成员作为抑癌基因，促进细胞周期阻滞和凋亡。FoxO 家族任何成员的表达上调，均会激活细胞周期抑制剂 p27 Kip1。Forkhead 转录因子也参与 TGF-β 介导的 p21 Cip1 上调，而这个上调过程可以被 PI3K 负调控 (7)。Forkhead 转录因子通过 Akt 在 Thr24、Ser256 和 Ser319 位点的磷酸化灭活时，增殖增加，导致出核和转录因子活性受到抑制 (8)。Forkhead 转录因子还可被 deacetylase sirtuin (SirT1) 抑制 (9)。 1.Anderson, M.J. et al.
