Pepper is able to enhance HBC's fluorescence more than 3,000-fold and exhibits bright fluorescence when expressed in living cells. HBC514 (2-10 μM) is altered flexibly by simple washing and staining by Pepper based CRISPR imaging in live HeLa cells (expressing sgRNA-Pep1 and dCas9).
