IKK gamma, also known as NEMO (NF-κB essential modulator), is a crucial regulatory subunit of the IκB kinase (IKK) complex. The IKK complex, which also includes the catalytic subunits IKKα and IKKβ, plays a central role in activating the NF-κB signaling pathway. IKK gamma itself does not possess kinase activity, but it is essential for the assembly and activation of the IKK complex. When cells are stimulated by various factors such as cytokines, bacterial products, or viral infections, the IKK complex is activated, leading to the phosphorylation and degradation of IκB proteins. This process releases NF-κB dimers, allowing them to translocate to the nucleus and regulate the expression of numerous genes involved in inflammation, immune responses, and cell survival. Mutations in the gene encoding IKK gamma can lead to severe immunodeficiencies and developmental disorders, highlighting its critical role in maintaining immune function and cellular homeostasis.
