BMS-986449 是一种 CELMoD 分子胶，通过诱导 Cereblon E3 泛素连接酶降解 Treg 特异性转录因子 Helios 和 Eos，从而重编程 Treg 细胞的免疫抑制表型。在激活状态下的人源 Treg 细胞中，BMS-986449 可在 8 天内显著降低 Helios 和 Eos 蛋白水平，促进 Granzyme B 等效应分子的表达，并上调炎症相关基因，展现出 Treg 向促炎功能转变的表型变化[1]。
In vivo(体内研究)
在携带人源 CRBN 的小鼠模型中，BMS-986449 每日口服可在多种肿瘤模型（CT26、MB49、MC38）中诱导明显的抗肿瘤效应，肿瘤抑制率分别为 75%、62%、42%，联合抗 PD-1 治疗后可进一步提升至 95%、97%、92%。此外，在食蟹猴中每日给药 0.1 mg/kg 可在外周 Treg 细胞中维持超过 80% 的 Helios 降解[1]。
