在与cathepsin L预孵育4小时后，SID 26681509的抑制作用增强（IC50: 1.0 nM）。确定SID 26681509为一种缓慢结合且缓慢可逆的竞争性抑制剂。通过对单步可逆性的瞬态动力学分析，得出抑制速率常数分别为kon = 24,000 M^-1s^-1和koff = 2.2 × 10^-5 s^-1（Ki = 0.89 nM）。通过基于实验推导的papain/CLIK-148 X射线晶体结构进行分子对接研究[1]。SID 26681509抑制papain和cathepsins B、K、S、V，经过一小时测定的IC50值介于618 nM至8.442 μM之间[1]。
