TNF-R2, or Tumor Necrosis Factor Receptor 2 (also known as CD120b), is a membrane-bound receptor primarily expressed on immune and endothelial cells. It binds to TNF-alpha, a key inflammatory cytokine, but transmits different signals than the related TNF-R1. TNF-R2 activation generally promotes cell survival, proliferation, and immunoregulatory functions. Detection of TNF-R2, particularly its soluble form (sTNF-R2) shed into the bloodstream, has significant clinical importance. Measuring sTNF-R2 levels via techniques like ELISA serves as a stable biomarker for overall TNF system activity. Elevated levels indicate chronic inflammatory and immune activation, correlating with disease severity in conditions like rheumatoid arthritis, sepsis, and certain cancers. Its detection aids in monitoring disease progression and therapeutic response, offering insights into the inflammatory state more reliably than fluctuating TNF-alpha itself.
