WNT5A and WNT5B are two highly homologous secreted glycoprotein ligands within the WNT signaling pathway family, playing critical yet distinct roles in embryonic development, tissue homeostasis, and disease pathogenesis. WNT5A, as the prototypical member of the non-canonical WNT signaling pathway, primarily activates β-catenin-independent signaling cascades—such as the WNT/Ca²⁺ pathway and the planar cell polarity pathway—by binding to Frizzled receptors and co-receptors like ROR1/ROR2, thereby regulating cell migration, adhesion, and polarity. It is essential for the normal formation of the skeleton, lungs, and heart during embryonic development; WNT5A knockout mice are nonviable due to severe developmental defects, and mutations in this gene can cause autosomal dominant Robinow syndrome in humans. In diseases such as cancer, the role of WNT5A is highly context-dependent: it can act as a tumor suppressor to inhibit tumor growth and metastasis (for instance, its loss of expression in breast cancer is often associated with poor prognosis), but it can also promote tumor aggressiveness under certain circumstances (such as its elevated expression in melanoma being linked to disease progression). In comparison, WNT5B shares approximately 80% amino acid sequence homology with WNT5A and also belongs to the WNT family, participating in developmental processes including cell differentiation and organ morphogenesis. However, its specific functions and regulatory mechanisms are relatively less studied, and it is known to play roles in processes such as muscle cell differentiation, skeletal system development, and angiogenesis.
