GTP cyclohydrolase 1 (GCH1) catalyzes the rate-limiting step of tetrahydrobiopterin (BH4) biosynthesis. Studies show that BH4 functions as a radical-trapping antioxidant and protects cells against ferroptosis independently of GPX4. In addition, dihydrofolate reductase (DHFR) is required to maintain BH4 levels by regenerating the compound (1,2). Furthermore, deficiency of spliceosomal GTPase Eftud2 in mouse cerebellar Purkinje cells (PCs) causes severe ferroptosis, PC degeneration, dyskinesia, and cerebellar atrophy. Ablation of Eftud2 in PCs downregulates Gch1 expression through mis-splicing of Atf4, activating ferroptosis (3).
