In vitro(体外研究)
RP-6306 (500 nM; 24h) treatment induced pan-γH2AX in HCC1569 breast cancer cell line indicates that tumor derived CCNE1 amplification also makes cells susceptible to DNA damage induction after PKMYT1 inhibition. RP-6306 treatment can selectively and accidentally activate CDK1 in CCNE1 overexpression cells, thus promoting early mitosis of cells undergoing DNA synthesis [2].
In vivo(体内研究)
In the ovarian xenograft model (OVCAR3) amplified by CCNE1, RP-6306 (15, 50, and 300 ppm; oral; daily; for 21 days) significantly reduced tumor growth in OVCAR3 in a dose-dependent manner [1].
