Suberoyl bis-hydroxamic Acid, an HDAC (histone deacetylase) inhibitor, inhibits HDAC1 (ID50 = 0.25 μM) and HDAC3 (ID50 = 0.3 μM). Suberoyl bis-hydroxamic Acid has been shown to induce differentiation of certain murine cells via blocking of HEXIM1 (hexamethylene bisacetamide) activity. Other experiments have shown Suberoyl bis-hydroxamic Acid to induce apoptosis in proliferative cells via induction of P21, P53, PUMA, and Bax gene expression. Additionally, the compound has been noted to activate Notch 1 signaling, which is associated with antiproliferative and apoptotic effects in certain medullary cells. Additional proteins induced by Suberoyl bis-hydroxamic Acid include caspase-3, poly ADP-ribose polymerase, Bcl-X(L), and Bad whose effects range from cellular growth, senescence and apoptosis.
