High performance sandwich TR-FRET immunoassay for semi-quantitative detection of Human Total STAT6 (both phosphorylated and non-phosphorylated) in cell lysates. This kit can be used to normalize data obtained with the Phospho-STAT6 (Y641) kit. Full assay validation can be found in the Technical Data Sheet. Detailed assay protocols are described in the Generic User Manual.

Stimulation of Phospho-STAT6 (Y641) in HeLa cells

Data show that treatment of HeLa cells with IL-4 stimulates phosphorylation of STAT6 at Y641, but does not affect the levels of total STAT6.

Total SAT6 (Y641) in HeLa cells-Time course of detection

Data show that robust S/B ratio and pharmacology can be obtained after 1h of incubation.

Inhibition of Phospho-STAT6 (Y641) in HeLa cells

Data show that treatment of HeLa cells with JAK Inhibitor 1 inhibits phosphorylation of STAT6 at Y641 by IL-4, but does not affect the levels of total STAT6.

Data show a linear relationship between lysate dilutions and TR-FRET ratio values.
