Sandwich TR-FRET immunoassay for semi-quantitative detection of Total AKT 1/2/3 (both phosphorylated and non-phosphorylated AKT1/2/3) in cell lysates. This kit can be used to normalize data obtained with the Phospho-AKT pan (S473) kit and the Phospho-AKT pan (T308) kit. Full assay validation can be found in the Technical Data Sheet. Detailed assay protocols are described in the Generic User Manual.

Data show that treatment of NIH3T3 cells with PDGF-AA stimulates phosphorylation of AKT pan at S473 but does not affect the levels of total AKT pan.
Data show that treatment of NIH3T3 cells with Wortmannin inhibits phosphorylation of AKT pan at S473 by PDGF-AA but does not affect the levels of total AKT pan.
Data show a linear relationship between lysate dilutions and TR-FRET ratio values.
