Sandwich TR-FRET immunoassay for semi-quantitative detection of Total Ribosomal Protein S6 (both phosphorylated and non-phosphorylated RPS6) in cell lysates. This kit can be used to normalize data obtained with the Phospho-RPS6 (S235/S236) kit and the Phospho-RPS6 (S240/S244) kit. Full assay validation can be found in the Technical Data Sheet. Detailed assay protocols are described in the Generic User Manual.

Data show that treatment of MCF7 cells with insulin stimulates phosphorylation of RPS6 at S235/S236, but does not affect the levels of total RPS6
Data show that treatment of MCF7 cells with LY294002 inhibits phosphorylation of RPS6 at S235/S236 by insulin, but does not affect the levels of total RPS6
Data show a linear relationship between lysate dilutions and TR-FRET ratio values
